Solid tumor metastases to bone cause significant morbidity. Bone is a unique microenvironment in which certain tumors thrive. Special features of the tumor cell and the bone drive a feed-forward cycle of tumor growth and abnormal bone remodeling. Preclinical and clinical evidence to support targeted therapy of molecular mediators of bone metastases will be discussed.
